Extraction of Bromelain

In order to get rid of the remnants of the culture, the plants are washed under running water. They are then put through a first extraction and run through a filter press to produce a juice that contains between 60 and 80 percent bromelain and is stored at 0 to 5 °C.

The resultant pulp, which naturally includes bromelain, is dissolved at a concentration of between 0.03 and 0.2 M, at a pH range of between 5.3 and 7.2, in an extraction buffer containing cold (0-5 ° C) potassium phosphate (K2PO4) to prevent denaturalization of the enzyme.

After allowing it to rest for 20 to 30 minutes, the mixture is run through a filter press to obtain a second extraction juice; the first juice and the second juice are then combined at 2000 to 5000 rpm for one to two minutes. Next, the mixture is centrifuged at 5000–10,000 rpm for 15-20 minutes at 0–4 °C to remove precipitate and recover the supernatant, which is then preserved at –80 to –2 °C. Finally, the extract is lyophilized for which said extract should contain at least 10-15% of solids.
